The literal answer is no. To do a variety of things with the measurements, yes.
Since there is no reason not to take measurements with a timing reference, why not do it?
 
ok thanks. In fact I continue to take measurements with acoustic timing inserted. But I wanted to ask you. In your experience, is the red curve or the black curve more musical?

No. You will need to measure each speaker separately to determine if the mic is perfectly centered. When the mic is perfectly centered the peak of each speaker’s impulse response will be at the same time. A timing reference will be required for the measurements.
