Calibrating the Interface

Dear all
It is not clear to me how to <<Connect the interface's line or headphone output directly to its line input>>.
My setup is the following:
MAC with soundcard connected USB
Soundcard output is toslink
Power Amplifier input is Toslink from soundcard
Power amplifier out is going to the loudspeakers
Soundcard input is via XLR to the calibrated dynaudio EMM6.

is it supposed to connect the speaker cable to the XLR input?

That would risk destroying the soundcard input if the level were too high. Safer to skip that part, but make sure monitoring is off on the soundcard so that the mic input is not being fed back to the soundcard output.
