If you only want to know how long sound takes to get from speaker to microphone use a tape measure to measure the distance and divide by the speed of sound, which is about 343 m/s in dry air at 20 C.
If you make a measurement with a loopback timing reference you will get the time taken from the signal leaving the interface to arriving back at the interface, assuming your mic is connected to the same interface. That may be longer then the time taken for the sound to travel from speaker to mic depending on what processing delays there are in the replay path.
